Sleep disruptions in middle age may signal Alzheimer's risk

A study of over 500 healthy adults found that more frequent brief awakenings during sleep were associated with higher genetic risk for Alzheimer's disease, but only in middle-aged participants. The findings suggest sleep measurements could serve as an early warning sign before memory problems develop. The research was published in the journal Sleep.
The study tracked over 500 healthy volunteers split into younger and middle-aged groups, calculating each person's genetic susceptibility using polygenic risk scoring, which condenses the combined effect of numerous genes into a single estimate. Researchers then compared these scores against various recorded sleep characteristics, including the frequency of brief brain-activity bursts known as micro-awakenings.
Notably, the association between these micro-awakenings and genetic risk emerged only among participants aged 50 to 69, not in the younger cohort. The locus coeruleus, a rice-grain-sized brainstem region governing wakefulness and sleep, drew particular attention. Researchers suggest certain sleep profiles might facilitate accumulation of Alzheimer's-related proteins, though the study's genetic risk scores remained low overall and cannot predict individual outcomes.
This research could eventually enable earlier identification of Alzheimer's vulnerability through routine sleep monitoring, potentially years before memory symptoms surface.
